尼日利亚菌素(Nigericin),来源于吸水链霉菌(Streptomyces hygroscopius)代谢产物的一种聚醚抗生素,常常用作钾离子载体,促进穿透生物膜的K+/H+交换,作用机制类似于缬氨霉素(货号:MS0049-10MG),其携带单价阳离子跨膜的特异性为:K+>Rb+>Cs+>>Na+。还可用作Pb2+离子载体,但对其他二价阳离子未表现出载运活性。作为一种有效的研究工具,能够激活ATPase酶活性,并且破坏线粒体膜电位。通过加强离子的膜扩散能力来杀死细菌。类似于ATP,尼日利亚菌素通过缝隙连接蛋白1(Pannexin-1)依赖性的方式,刺激胞内K+外流,诱导NLRP3炎症小体组装,凋亡相关蛋白Caspase-1成熟,进而刺激IL-1β的加工和释放。

实验方法(Assay):To evaluate the effect of ΔpH on alkali tolerance, growth experiments using four strains of lactobacilli and E. hirae JCM 8729T were conducted in the presence or absence of nigericin at pH values between 6.5 and 9.0. Nigericin in ethanol solution was added at a final concentration of 7 μM. Control cultures received 0.1% (vol/vol) ethanol. Specific growth rates were calculated during the exponential growth phase as described above. |

动物模型(Animal Model):Nude mice injected with S19 cells
实验方法(Assay):Mice are randomly divided into four groups with six animals each (control, DDP, Nigericin and DDP combined with Nigericin). DDP (2.5 mg/kg) is injected intraperitoneally for five continuous days and nigericin (4 mg/kg) is administrated intraperitoneally every two days.
