Background

Interleukin-12 (IL-12), also known as NKSF or CLMF, is a pleiotropic cytokine originally identified in the medium of activated human B lymphoblastoid cell lines. The p40 subunit of IL-12 has been shown to have extensive amino acid sequence homology to the extracellular domain of the human IL-6 receptor while the JP30 subunit shows distant but significant sequence similarity to IL-6, G-CSF, and chicken MGF. These observations have led to the suggestion that IL-12 might have evolved from a cytokine/soluble receptor complex. Human and murine IL-12 share 70% and 60% amino acid sequence homology in their p40 and JP30 subunits, respectively. IL-12 apparently shows species specificity with human IL-12 reportedly showing minimal activity in the murine system.

Background

Interleukin-11 (IL-11) is encoded by the IL11 gene and is a member of IL-6 superfamily, distinguished based on their use of the common co-receptor gp130. It is a multifunctional cytokine first isolated from bone marrow-derived stromal cells. IL-11 receptor activation requires formation of a complex of two IL-11 molecules with two molecules of the ligand-binding IL-11 Rα subunit and two molecules of the expressed cell signaling β subunit, gp130. The functions of IL-11 are that it directly stimulates the proliferation of hematopoietic stem cells and megakaryocyte progenitor cells, and induces megakaryocyte maturation resulting in increased platelet production.

Background

Interleukin-10 (IL-10), also known as cytokine synthesis inhibitory factor (CSIF), is the charter member of the IL-10 family of α-helical cytokines that also includes IL-19, IL-20, IL-22, IL-24, and IL-26/AK155. IL-10 is secreted by many activated hematopoietic cell types as well as hepatic stellate cells, keratinocytes, and placental cytotrophoblasts. Whereas human IL-10 is active on mouse cells, mouse IL-10 does not act on human cells. IL-10 is a 178 amino acid molecule that contains two intrachain disulfide bridges and is expressed as a 36kDa noncovalently associated homodimer. The IL-10 dimer binds to two IL-10Rα/IL-10R1 chains, resulting in recruitment of two IL-10Rβ/IL-10R2 chains and activation of a signaling cascade involving JAK1, TYK2, and STAT3. IL-10Rβ does not bind IL-10 by itself but is required for signal transduction. IL-10 is a critical molecule in the control of viral infections and allergic and autoimmune inflammation. It promotes phagocytic uptake and Th2 responses but suppresses antigen presentation and Th1 proinflammatory responses.

Background

Interleukin-9 (IL-9) is encoded by the IL9 gene and produced by T-cells and specifically by CD4+ helper cells. IL-9 was originally identified as a cytokine found in the conditioned medium of a human T cell leukemia virus type I (HTLVI) transformed T cell line. It functions through the IL-9 receptor, which activates different signal transducer and activator (STAT) proteins and thus connects this cytokine to various biological processes. IL-9 can support the growth of IL-2 independent and IL-4 independent helper T-cells. Human IL-9 has approximately 56% amino acid sequence identity with murine IL-9. The gene encoding this cytokine has been identified as a candidate gene for asthma. Background

Interleukin-7 (IL-7) is encoded by the IL7 gene and secreted by stromal cells in the red marrow and thymus. It binds to the IL-7 receptor, a heterodimer consisting of IL-7 receptor alpha and IL-2 receptor gamma chain. Interleukin-6 (IL-6) is an interleukin that in humans is encoded by the IL-6 gene and acts as both a pro-inflammatory and antiinflammatory cytokine. The IL-5 gene is located on chromosome 5 in humans, in close proximity to the genes encoding IL-3, IL-4, and granulocytemacrophage colony-stimulating factor (GM-CSF), which are often co-expressed in TH2 cells. Interleukin-4 (IL-4) is a pleiotropic cytokine that induces differentiation of naive helper T cells (Th0 cells) to Th2 cells and produced by mast cells, activated T cells and bone marrow stromal cells. IL-2 is a powerful immunoregulatory lymphokine produced by T-cells in response to antigenic or mitogenic stimulation. Background

Interleukin-1 receptor antagonist (IL-1RA) is a member of the IL-1 family. IL-1RA is secreted by various types of cells including immune cells, epithelial cells, and adipocytes, and is a natural inhibitor of the pro-inflammatory effect of IL-1β. This protein inhibits the activities of interleukin 1, alpha (IL-1α) and interleukin 1, beta (IL-1β), and modulates a variety of interleukin 1 related immune and inflammatory responses. IL-4 and IL-13 have been shown to amplify the stimulatory effect of IL1-beta on the production of soluble and intracellular forms of IL1-RA. The regulated expression of IL-1RA in various cell types has been shown to be influenced by cytokines. In synovial fibroblasts the synthesis of IL-1RA is markedly enhanced by IL-1, TNF-α, or PDGF. The protein shows 26% amino acid homology to IL-1β and 19% homology to IL-1α.

胰蛋白酶(胰酶,Trypsin),CAS:9002-07-7,来源于胰腺的一种丝氨酸蛋白酶,由223个氨基酸残基组成的单链多肽,底物特异性是带正电荷的赖氨酸和精氨酸侧链。胰酶主要切割赖氨酸和精氨酸羧基端,当两者之一紧随为脯氨酸的情况除外。另外,当切割位点任一边紧邻酸性残基,胰酶水解速率也会减缓。

使用方法

1. 产品溶解

储存液制备:溶于1mM HCl(pH 3)配制母液,充分溶解,-20℃分装冻存,至少稳定保存~1年。加入Ca2+(20mM)能延缓胰蛋白酶的自溶和维持胰蛋白酶在溶液内的稳定性。

2. 产品用法

使用时,将储存液稀释在50mM NH4HCO3, pH 7.8或其它的合适缓冲液内。

胰蛋白酶:目的蛋白=1:50~1:1000,最适pH 7.0-9.0,最适温度:25-35℃。需注意,本品的活性受丝氨酸蛋白酶抑制剂比如:PMSF的抑制,金属离子螯合剂比如:EDTA的抑制。

注意事项

1)胰蛋白冻干粉产品常遇到的活性定义及其转换,

1 BAEE Unit:在25℃,pH 7.6的条件下,以BAEE为底物,3.2 ml的反应体系中,A253吸光值每分钟会发生0.001的变化即为一个酶活单位。

1 TAME Unit: 在25℃,pH 8.2,0.001M Ca2+条件下,每分钟水解1μmol p-toluenesulfonyl-L-arginine methyl ester(TAME)的用量。

1 USP Unit:在指定条件下,每分钟引起吸光值发生0.003的变化的样品活性。

重组赖氨酰内切酶(Recombinant Lysyl Endopeptidase, rLys-C)属于丝氨酸蛋白酶,特异性切割赖氨酸残基羧基端的肽键。Lys-C广泛用于各种生物技术过程中,如:蛋白质酶解生产多肽;GLP-1类药物制备等。生物活性受 DFP、PMSF、TLCK 抑制,不被TACK抑制。

本品为大肠杆菌重组表达所得,无外源性的病毒污染,生产过程中不使用任何动物源的原料。基于稳定的重组生产工艺,可批量生产,批次间几乎无差异,质量稳定,且纯度高,低内毒素水平。以冻干粉的形式提供,酶活力≥0.5AU /mg powder。rLys-C适用于重组胰岛素及类似物生产、GLP-1类似物生产、蛋白质质谱、测序、肽图谱分析;以及小分子蛋白或多肽生产等。

保存与运输方法

使用方法

  1. 产品溶解

储存液制备:溶于20-50mM Tris-HCl(pH 8.5-9.5)配制母液,充分溶解,-20℃分装冻存,数月稳定保存。配制好的储存液可承受反复冻融~5次。

  1. 产品用法

使用时,将储存液加入样本溶液,按照酶:蛋白=(1-100AU):1g的比例来优化加量,最适pH:9.0-9.5,最适温度30-37℃,反应时间2~24h。

【注意】:如果样本所含的无机盐过高(≥0.1M),会影响酶活。建议对样本进行脱盐后再进行酶解反应。若不能进行脱盐,建议通过增加酶量且延长孵育时间来达到所需效果。

胰岛素(Insulin)是胰腺内胰岛β细胞分泌的一种由α和β双链构成的多肽激素,由51个氨基酸组成,分子量约5800 Da。两条链由两对链间二硫键连接,另外α链自身还含有一对链内二硫键。胰岛素是目前已知的机体内唯一能降低血糖的激素,不仅负责调控细胞对葡萄糖、氨基酸和脂肪酸的摄入,利用和储存,而且抑制糖原,蛋白质和脂肪的降解发生。

不同物种胰岛素功能大体相同,只是氨基酸组成上的差异。人胰岛素结构见下:

常见问题

1. 选择什么溶剂来溶解胰岛素干粉?

胰岛素中性环境几乎不溶解,但可迅速溶解于稀酸或稀碱,测试数据表明胰岛素溶于0.01M HCl的浓度可达20 mg/ml。

2. 胰岛素的母液如何配制和保存?

将胰岛素溶于稀醋酸(1%)或稀盐酸(pH 2-3)配制1-10mg/ml的储存液,用低蛋白吸附滤膜对配制好的溶液进行过滤除菌,按照单次使用剂量,分装冻存于-20℃,避免反复冻融。至少6个月稳定。

【注意】:胰岛素虽可溶于125mM碳酸氢钠溶液,但不建议制备碱性储存液,原因在于高pH会导致发生脱酰胺和聚集反应的速率提高。胰岛素溶液不可进行高压灭菌处理。

3. 胰岛素的应用和使用浓度?

胰岛素中重组人胰岛素、牛胰岛素或猪胰岛素可用作细胞培养添加物。体外研究胰岛素长期被认为是用来调控大部分细胞生长和分化的关键因子。常用使用浓度范围是1-10μg/ml,主要根据细胞类型来决定。另外,对于要求无蛋白细胞培养体系,可将胰岛素固定在聚苯乙烯培养皿表面,以提高细胞的粘附性。
