Background

Interleukin-36 (IL-36) is a pro-inflammatory cytokine which plays an important role in the pathophysiology of several diseases. IL-36α, IL-36β, and IL-36γ (formerly IL-1F6, IL-1F8, and IL-1F9) are IL-1 family members that signal through the IL-1 receptor family members IL-1Rrp2 (IL-1RL2) and IL-1RAcP. IL-36γ is secreted when transfected into 293-T cells and it could constitute part of an independent signaling system analogous to interleukin-1 alpha (IL-1A), beta (IL-1B) receptor agonist and interleukin-1 receptor type I (IL-1R1). Furthermore, IL-36γ also can function as an agonist of NF-kappa B activation through the orphan IL-1-receptor-related protein 2. IL-33, encoded by IL-33 gene located on the Chr.9 in humans, is a 30kDa proinflammatory protein belonging to the IL-1 superfamily and it shares less than 20%a.a. sequence identity with other members. IL-33 secreted by high endothelial venules at high levels, which is found in tonsils, peyer patches and mesenteric lymph nodes, but not in placenta. It is upregulated in arterial smooth muscle, dermal fibroblasts, and keratinocytes following IL1α or IL1β stimulation. Background

IL-17B, also named IL20, NIRF and ZCYTO7, is belonging to the IL-17 family and it stimulates the release of tumor necrosis factor alpha and IL-1-beta from the monocytic cell line THP-1. IL-17B mRNA is expressed in adult pancreas, small intestine, and stomach. The IL-17 family is comprised of at least six proinflammatory cytokines that share a conserved cysteine-knot structure but diverge at the N-terminus. IL-17 family members are glycoproteins secreted as dimers that induce local cytokine production and recruit granulocytes to sites of inflammation. IL-17B binds the IL-17B receptor, but not the IL-17 receptor; it is most homologous with IL-17D, which is expressed by resting CD4+ T cells and CD19+ B cells. This cytokine was reported to stimulate the release of TNF alpha (TNF) and IL1 beta (IL1B) from a monocytic cell line. Immunohistochemical analysis of several nerve tissues indicated that this cytokine is primarily localized to neuronal cell bodies.

Human Interleukin-16 (IL-16) is also called lymphocyte chemoattractant factor (LCF) and it has three isoforms which are cleaved from pre-Interleukin-16. IL-16 is mostly secreted by lymphocytes, epithelial cells, eosinophils, and CD8+ T cells. It has many functions including induction of the IL-2Rα on T cells, suppression of human immunodeficiency virus (HIV) replication, inhibition of T-cell antigen receptor/CD3 mediated T-cell stimulation in mixed lymphocyte reactions and so on. Background

Human Interleukin-13 (IL-13) is expressed by the IL13 gene located on the chromosome 5. IL-13 is a 132 amino acid protein containing a proposed 20 amino acid signal peptide, and shares approximately 30% amino acid sequence homology to human IL-4. Additionally, the two cytokines exhibit overlapping biological activities. Human IL-13 is produced by activated Th0, Th1-like Th2-like and CD8 T cells. Similarly to IL-4, IL-13 has multiple effects on the differentiation and functions of monocytes/macrophages. IL-13 can suppress the cytotoxic functions of monocytes/macrophages.
IL-13 variant is a natural variant of IL-13 and has a Q instead of R112 of wild type, which associated with increased risk for asthma development and at homozygosity associated with higher levels of serum total IgE in some allergic rhinitis patients.
