酵母菌的质粒筛选通常基于营养缺陷型突变菌株的使用而开展,这些菌株在缺乏某种特定培养基组分(比如氨基酸,嘌呤或嘧啶)的情况下无法生长。当转化含有突变基因的质粒后使得阳性转化克隆能够在缺乏必需营养组分的培养基内生长,正因如此,省却成分培养基添加剂(DO medium supplements)能用来筛选转化的酵母培养物。

本品为缺乏蛋氨酸(Met)的省却成分培养基添加剂,与Minimal SD base联合使用,配制成单组分缺陷的酵母菌选择性培养基,俗称单缺培养基(一缺培养基),主要用于单质粒酵母转化筛选。可用在外源基因在酵母中的表达、异源基因功能互补、酵母单杂交系统和酵母双杂交系统初始质粒转化。

配制方法(Formulation):fMLP was prepared at a concentration of 1 μg/μL in a solution of 25 % DMSO and 75 % PBS. Vehicle control was a 5 μL solution of 25 % DMSO and 75 % PBS.

实验方法(Assay):12-week-old C57BL/6J mice received local intra-articular injection of fMLP or vehicle control at the right-knee joint. Mice were anaesthetised using 2 % isoflurane. A total of 5 μL of fMLP solution or vehicle control was injected into the right hindlimb knee joint using a 5 μL Hamilton® syringe and 27-Gauge needle, as previously described. The dosing of 5 μg of fMLP was chosen based on its maximum solubility in vehicle solution and maximum volume of solution that could be injected into the knee-joint space.

Each sample (0.25 ml) contained 0.5 × 106 neutrophils with 0.8 μM luminol. The reaction to fMLP (10 μM final concentration) or TNF-α (5 ng/ml final concentrations) was initiated by automated injection to ensure an immediate mixing and measurement.

产品描述

酵母菌的质粒筛选通常基于营养缺陷型突变菌株的使用而开展,这些菌株在缺乏某种特定培养基组分(比如氨基酸,嘌呤或嘧啶)的情况下无法生长。当转化含有突变基因的质粒后使得阳性转化克隆能够在缺乏必需营养组分的培养基内生长,正因如此,省却成分培养基添加剂(DO medium supplements)能用来筛选转化的酵母培养物。

本品为缺乏蛋氨酸(Met)/色氨酸(Trp)的省却成分培养基添加剂,与Minimal SD base联合使用,配制成双组分缺陷的酵母菌选择性培养基,俗称双缺培养基(二缺培养基),用于双质粒酵母转化筛选。可用在外源基因在酵母中的表达、异源基因功能互补、酵母单杂交系统和酵母双杂交系统初始质粒转化免疫共沉淀、Y187和AH109酵母交配实验、接合型二倍体筛选。

NPM 3 Network Particulate Monitor

The Met One Instruments, Inc. NPM 3 is a forward light scatter laser nephelometer particulate monitor which automatically measures real-time airborne TSP, PM10, or PM2.5 particulate concentration levels.

The NPM 3 features improved reliability and lower maintenance. Each unit uses a serial cable that allows the device to connect to a laptop, data logger, or CCS+ Comet Cloud Modem. The real-time data is accessible for display, logging, review, and reporting instantly or later time.

The monitor contains a 5-mW diode laser operating at 670 nm wavelength.  A protective optical housing fully encapsulates the laser beam and optics system within the device.

Data from the NPM 3 can be managed using the supplied copy of Met One’s user-friendly, Windows-based, Comet™ communications terminal software.   Comet allows the user to view real-time data and log data to a data file on a computer in .csv format.

The NPM 3 is calibrated using 0.6-micron latex microspheres. These provide an extremely consistent calibration but do not generally match the characteristics of all ambient particulate. A K Factor (multiplier) must be established using a collocated trusted reference source (such as a BAM 1020) for good accuracy and correlation. The default factory K Factor is 1.0.

The K Factor is only valid at the same site and for the same particulate type. If the local particulate source changes, the K Factor may need to be adjusted.

The unit is supplied with a TSP head, Comet software, a mounting bracket, & a hose clamp.

PARTAC Tactical Particulate Profiler

Met One’s PARTAC Tactical Particulate Profiler (P/N 102686) is a robust size-selective particulate sampler used as an NBC agent alarm trigger, an ambient monitor for public and workplaces, and as a ground-truth reference for the verification of standoff particulate detectors. The PARTAC simultaneously monitors airborne particle counts in up to 8 user-selectable channels.

The PARTAC can be programmed to detect the particulate sizes of toxic NBC agents proven to be aspirated deeply into the lungs. Measuring the particulate counts in real-time, the PARTAC can be used to trigger alarms and activate additional samplers and instrumentation. Its compact size, low power consumption, and continuous duty capability allow it to perform this duty continuously in all environments.

As an ambient monitor, it provides the user with a real-time count of particulates in public or workplaces. Its low noise operation allows it to be placed in work environments without disturbing the personnel in the area being monitored. The PARTAC can also be placed in areas where work is being contemplated to measure the background levels before work begins.

The PARTAC sensor uses a laser diode to illuminate the individual particles drawn into the PARTAC through the sampling inlet by the integral pump and mass flow control system. The laser light scattered at right-angles to the incident light beam is collected and focused onto a photodetector using a high-grade elliptical reflector. The signal strength is detected by state-of-the-art electronics and converted to a set of size values for each detected particle. These counts are classified into the size ranges set by the customer prior to installation, at a rate of up to once per second.

BAM-1022 Beta Attenuation Mass Monitor

The Met One Instruments, Inc. BAM 1022 Real-Time Portable Beta Attenuation Mass Monitor continuously measures the mass concentration of ambient particulate matter collected onto glass filter tape with a time resolution of one minute. The BAM 1022 employs an in-line sampling geometry in which the attenuation of beta rays across filter media is measured and particulate matter is sampled simultaneously. This allows ambient sampling to occur for virtually 60 minutes each hour. It also allows for beta ray measurements to be made for virtually the entire hour thereby improving sensitivity and time resolution.

The BAM 1022 employs a unique “in-situ” sampling technique where the beta measurement is kept at a nearly fixed temperature above ambient conditions, thereby minimizing measurement error due to loss of semi-volatile particulate material or excessive moisture in the sample stream. Highly accurate measurements are made without having to employ expensive Nafion dryers, thereby increasing reliability and decreasing operating costs. The integrated shelter does not employ air conditioning, nor does the sampling system employ flow splitters. This results in easier service, enhanced reliability, and lower power consumption.

The BAM 1022 comes integrated into its own lightweight equipment shelter with an easily detachable pump box making it ideal for installation in areas where no permanent structure or walk-in shelter is available. Power consumption is modest as no air conditioning is required. No component weighs more than 40 lb. meaning that it is easily transportable by one person. The entire system may be set up and put into operation in less than ten minutes.

Particulate Sampler E-FRM Reference

Met One Instruments’ E-FRM is a single channel particulate sampler approved as a US-EPA reference method for PM2.5, PM10, PM10-2.5. Configured with an alternate PM2.5 cyclone, the E-FRM may also be operated as a US-EPA designated equivalent method for PM2.5. For ease of shipping, the E-FRM is nested in a compact package. Met One Instruments’ Industrial Weather Stations are configured to provide plant operators with the weather data needed for operations, regulatory compliance, safety and emergency response. The systems use reliable and accurate sensors to measure the weather variables of interest. Depending on output/input requirements, the data logger outputs either an analog voltage or digital RS-232 / RS-485 in ASCII or MODBUS format, or 4 -20 mA current loop for connection to the plant’s DCS, PLC’s or SCADA system. Met One Instruments, Inc.’s Model GT-324 particle counter is a full-featured handheld particle counter that provides the functionality of a bench unit in a portable package. A fully integrated RH/Temp sensor immediately provides data to your displayed and logged data. A convenient unit for compliance, the user interface is simple with the multifunction rotary dial.
