溶解性:易溶于水,但其水溶液在室温下极不稳定,可在半小时后分解为气体而挥发掉,故需现用现配。溶于较低度醇和酮,不溶于极性的有机溶剂。
生物活性:本品为亚硝脲类抗生素,主要用于胰岛细胞癌(β细胞或非β细胞癌),对类癌肿瘤、霍奇金病、结肠癌及肝癌等变有一定疗效。它是一种有效的 DNA 甲基化试剂,作用于 HL60,K562 和 C1498 细胞,IC50 分别为11.7,904和1024μg/mL。实验室常用于糖尿病模型的复制。
用途:一种含N-亚硝基的化合物,在胰腺胰岛充当一氧化氮的供体;诱导产生糖尿病动物模式的胰岛素分泌细胞的死亡。高效DNA甲基化试剂,能诱发染色体断裂。对表达GLUT2葡萄糖转运体(GLUT2 glucose transporter)的神经内分泌肿瘤细胞系具细胞毒性。医学研究上链脲佐菌素用于诱导Type1糖尿病。医学研究上链脲佐菌素用于诱导Type1糖尿病。

产品介绍 Immunoglobulin G (IgG), is one of the most abundant proteins in serum with normal levels between 8-17 mg/mL in adult blood. IgG is important for our defence against microorganisms and the molecules are produced by B lymphocytes as a part of our adaptive immune response. The IgG molecule has two separate functions; to bind to the pathogen that elicited the response and to recruit other cells and molecules to destroy the antigen. The variability of the IgG pool is generated by somatic recombination and the number of specificities in an individual at a given time point is estimated to be 1011 variants.

This gene encodes one of six different actin proteins. Actins are highly conserved proteins that are involved in cell motility, structure, and integrity. This actin is a major constituent of the contractile apparatus and one of the two nonmuscle cytoskeletal actins. [provided by RefSeq, Jul 2008].

Function:
Actins are highly conserved proteins that are involved in various types of cell motility and are ubiquitously expressed in all eukaryotic cells.

Subunit:
Polymerization of globular actin (G-actin) leads to a structural filament (F-actin) in the form of a two-stranded helix. Each actin can bind to 4 others. Identified in a mRNP granule complex, at least composed of ACTB, ACTN4, DHX9, ERG, HNRNPA1, HNRNPA2B1, HNRNPAB, HNRNPD, HNRNPL, HNRNPR, HNRNPU, HSPA1, HSPA8, IGF2BP1, ILF2, ILF3, NCBP1, NCL, PABPC1, PABPC4, PABPN1, RPLP0, RPS3, RPS3A, RPS4X, RPS8, RPS9, SYNCRIP, TROVE2, YBX1 and untranslated mRNAs. Component of the BAF complex, which includes at least actin (ACTB), ARID1A, ARID1B/BAF250, SMARCA2, SMARCA4/BRG1, ACTL6A/BAF53, ACTL6B/BAF53B, SMARCE1/BAF57 SMARCC1/BAF155, SMARCC2/BAF170, SMARCB1/SNF5/INI1, and one or more of SMARCD1/BAF60A, SMARCD2/BAF60B, or SMARCD3/BAF60C. Oxidation of Met-44 by MICALs (MICAL1, MICAL2 or MICAL3) to form methionine sulfoxide promotes actin filament depolymerization. Methionine sulfoxide is produced stereospecifically, but it is not known whether the (S)-S-oxide or the (R)-S-oxide is produced.

DISEASE:
Defects in ACTA1 are the cause of nemaline myopathy type 3 (NEM3) [MIM:161800]. A form of nemaline myopathy. Nemaline myopathies are muscular disorders characterized by muscle weakness of varying severity and onset, and abnormal thread-or rod-like structures in muscle fibers on histologic examination. The phenotype at histological level is variable. Some patients present areas devoid of oxidative activity containg (cores) within myofibers. Core lesions are unstructured and poorly circumscribed.
Defects in ACTA1 are a cause of myopathy congenital with excess of thin myofilaments (MPCETM) [MIM:161800]. A congenital muscular disorder characterized at histological level by areas of sarcoplasm devoid of normal myofibrils and mitochondria, and replaced with dense masses of thin filaments. Central cores, rods, ragged red fibers, and necrosis are absent.

The secondary antibody used for 40 min at room temperature. Acquisition of 20,000 events was performed.

Isotype Control Antibody: Rabbit IgG/FITC(orange) ,used under the same conditions.
Protocol
The cells were fixed with 2% paraformaldehyde (10 min) and then permeabilized with ice-cold 90% methanol for 30 min on ice. The cells were washed twice with 1 X PBS. The cells were incubated in 1 X PBS containing 0.5% BSA + 1 0% goat serum (15 min) to block non-specific protein-protein interactions followed by the incubated with antibody (JP-0061R/FITC, 1μg /1×10^6 cells) for 30 min on ice. Acquisition of 20,000 events was performed.

产品介绍 All cells within tissues are surrounded by an extracellular matrix (ECM) giving the tissues shape and structure. The ECM is constantly being remodeled and constant communication is maintained between cells through this matrix. Secreted proteins, termed matrix metalloproteinases (MMPs), are involved in the modulation of cell matrix interactions. MMPs are Zn(2+) binding endopeptidases that degrade various components of the ECM. MMPs are enzymes implicated in normal and pathologic tissue remodeling processes, wound healing, angiogenesis, and tumor invasion. These enzymes are very potent when active, and are associated with extracellular space inhibitors called TIMPs (tissue inhibitors of matrix metalloproteinases). TIMPs have been shown to block tumor cell invasion suggesting that they act as metastasis suppressor genes.

Function:
May play an essential role in local proteolysis of the extracellular matrix and in leukocyte migration. Could play a role in bone osteoclastic resorption. Cleaves KiSS1 at a Gly-|-Leu bond. Cleaves type IV and type V collagen into large C-terminal three quarter fragments and shorter N-terminal one quarter fragments. Degrades fibronectin but not laminin or Pz-peptide.

Subunit:
Exists as monomer or homodimer; disulfide-linked. Exists also as heterodimer with a 25 kDa protein. Macrophages and transformed cell lines produce only the monomeric form. Interacts with ECM1.

Produced by normal alveolar macrophages and granulocytes.

Post-translational modifications:
Processing of the precursor yields different active forms of 64, 67 and 82 kDa. Sequentially processing by MMP3 yields the 82 kDa matrix metalloproteinase-9.
N- and O-glycosylated.

DISEASE:
Defects in MMP9 may be a cause of susceptibility to intervertebral disc disease (IDD) [MIM:603932]; also known as lumbar disk herniation (LDH). IDD is one of the most common musculo-skeletal disorders and the predominant cause of low-back pain and unilateral leg pain.
Defects in MMP9 are the cause of metaphyseal anadysplasia type 2 (MANDP2) [MIM:613073]. Metaphyseal anadysplasia consists of an abnormal bone development characterized by severe skeletal changes that, in contrast with the progressive course of most other skeletal dysplasias, resolve spontaneously with age. Clinical characteristics are evident from the first months of life and include slight shortness of stature and a mild varus deformity of the legs. Patients attain a normal stature in adolescence and show improvement or complete resolution of varus deformity of the legs and rhizomelic micromelia.

产品介绍 Beclin 1 is the first identified mammalian gene to mediate autophagy and also has tumor suppressor and antiviral function. Autophagy, a process of bulk protein degradation through an autophagosomic lysosomal pathway, is important for differentiation, survival during nutrient deprivation, and normal growth control, and is often defective in tumor cells. Beclin 1 was originally isolated in a yeast two hybrid screen to identify Bcl 2 binding partners and maps to a tumor susceptibility locus on human chromosome 17q21 that is frequently monoallelically deleted in human breast, ovarian and prostate cancer. Beclin 1 encodes an evolutionarily conserved 52kDa coiled coil protein that is expressed in human muscle, epithelial cells and neurons.

Function:
Plays a central role in autophagy. Required for the abcission step in cytokinesis. May play a role in antiviral host defense. Protects against infection by a neurovirulent strain of Sindbis virus.

Subunit:
Interacts with GOPC and GRID2. Interacts with AMBRA1. Forms a complex with PIK3C3 and PIK3R4 and either UVRAG and KIAA0226/Rubicon, or with ATG14. UVRAG and ATG14 form mutually exclusive complexes through direct competition for BECN1-binding. The complex containing ATG14 up-regulates autophagy, while the one containing Rubicon down-regulates autophagy. Interacts with BCL2 and BCL2L1. Interacts with VMP1. Interacts with PIK3CB. Interacts with DAPK1. Interacts with human cytomegalovirus/HHV-5 protein TRS1.

Subcellular Location:
Golgi apparatus, trans-Golgi network membrane; Peripheral membrane protein. Note=Interaction with ATG14 promotes translocation to autophagosomes. Expressed in dendrites and cell bodies of cerebellar Purkinje cells.

Tissue Specificity:
Ubiquitous.

Post-translational modifications:
Phosphorylation at Thr-119 by DAPK1 reduces its interaction with BCL2 and BCL2L1 and promotes induction of autophagy.
Ubiquitinated. Deubiquitinated by USP10 and USP13, leading to stabilize the PIK3C3/VPS34-containing complexes.
