Background

Interleukin-7 (IL-7) is encoded by the IL7 gene and secreted by stromal cells in the red marrow and thymus. It binds to the IL-7 receptor, a heterodimer consisting of IL-7 receptor alpha and IL-2 receptor gamma chain. IL-7 stimulates the differentiation of hematopoietic stem cells into lymphoid progenitor cells and also stimulates proliferation of B cells, T cells and NK cells. Murine IL-7 has approximately 65% amino acid sequence identity with human IL-7 and both proteins exhibit cross-species activity. IL-7 as an immunotherapy agent has been examined in many human clinical trials for various malignancies and during HIV infection.

Interleukin-6 (IL-6) is an interleukin that in humans is encoded by the IL-6 gene and acts as both a pro-inflammatory and antiinflammatory cytokine. It is secreted by T cells and macrophages to stimulate immune response. Furthermore, It plays an essential role in the final differentiation of B-cells into Ig-secreting cells involved in lymphocyte and monocyte differentiation. It also induces myeloma and plasmacytoma growth and induces nerve cells differentiation acts on B-cells, T-cells, hepatocytes, hematopoietic progenitor cells and cells of the CNS. Background

The IL-5 gene is located on chromosome 5 in humans, in close proximity to the genes encoding IL-3, IL-4, and granulocytemacrophage colony-stimulating factor (GM-CSF), which are often co-expressed in TH2 cells. Through binding to the IL-5 receptor, IL-5 stimulates B cell growth and increases immunroglobulin secretion. It is also a key mediator in eosinophil activation. Interleukin-5 has long been associated with the cause of several allergic diseases including allergic rhinitis and asthma. IL-5 is a 115-amino acid (in human, 133 in the mouse) -long TH2 cytokine that is part of the hematopoietic family. Unlike other members of this cytokine family (namely IL-3 and GM-CSF), this glycoprotein in its active form is a homodimer.

Interleukin-4 (IL-4) is a pleiotropic cytokine that induces differentiation of naive helper T cells (Th0 cells) to Th2 cells and produced by mast cells, activated T cells and bone marrow stromal cells. Background

Interleukin-3 (IL-3) is an interleukin, a type of biological signal (cytokine) which is encoded by the IL-3 gene located on chromosome 5 and produced primarily by activated T cells beside human thymic epithelial cells, activated murine mast cells, murine keratinocytes and neurons/astrocytes. The protein acts in hematopoiesis by controlling the production, differentiation, and function of 2 related white cell populations of the blood, the granulocytes and the monocytes-macrophages. Background

IL-2 is a powerful immunoregulatory lymphokine produced by T-cells in response to antigenic or mitogenic stimulation. It is expressed by CD4+ and CD8+ T cells, γδ T cells, B cells, dendritic cells and eosinophils. Background

Interleukin-1 receptor antagonist (IL-1RA) is a member of the IL-1 family. IL-1RA is secreted by various types of cells including immune cells, epithelial cells, and adipocytes, and is a natural inhibitor of the pro-inflammatory effect of IL-1β. This protein inhibits the activities of interleukin 1, alpha (IL-1α) and interleukin 1, beta (IL-1β), and modulates a variety of interleukin 1 related immune and inflammatory responses. IL-4 and IL-13 have been shown to amplify the stimulatory effect of IL1-beta on the production of soluble and intracellular forms of IL1-RA. The regulated expression of IL-1RA in various cell types has been shown to be influenced by cytokines. In synovial fibroblasts the synthesis of IL-1RA is markedly enhanced by IL-1, TNF-α, or PDGF. The protein shows 26% amino acid homology to IL-1β and 19% homology to IL-1α.

The protein encoded by this gene is a secreted cytokine that is important for the proliferation of T and B lymphocytes. The receptor of this cytokine is a heterotrimeric protein complex whose gamma chain is also shared by interleukin 4 (IL4) and interleukin 7 (IL7). The expression of this gene in mature thymocytes is monoallelic, which represents an unusual regulatory mode for controlling the precise expression of a single gene. The targeted disruption of a similar gene in mice leads to ulcerative colitis-like disease, which suggests an essential role of this gene in the immune response to antigenic stimuli. ◆原理

　　细胞死亡后会释放一类特殊的蛋白质 Alarmin（暂译为警报素），通过检测 Alarmin 可识别组织创伤。然而在抗病毒免疫系统中，Alarmin 所起的作用不是很明确。最近瑞士日内瓦大学和德国柏林夏里特医科大学研究人员发现在脾脏和淋巴结中病毒的复制可导致受感染的细胞死亡。这些坏死的细胞可释放出 Alarmin，其中主要的是白介素-33（IL-33），它可通过 ST2 受体激活 CD8+ T（CTLs）细胞，对病毒感染作出反应。研究发现缺乏 IL-33 的小鼠，CTL 对病毒感染的免疫反应也较低。此项研究具有里程碑意义，因为提出了一个新的免疫机制：坏死细胞会释放出 Alarmin 动员杀伤性Ｔ细胞应对感染，在未来对抗艾滋、丙型肝炎而开发新的疫苗，对于癌症治疗有重要作用。

白细胞介素-6（IL-6）是调节免疫系统和造血系统的重要细胞因子。人IL-6由T细胞、肥大细胞、单核细胞、巨噬细胞、成纤维细胞、内皮细胞、角质形成细胞及许多肿瘤细胞系表达。在急性期反应、对损伤和炎症的反应中起作用。 人IL-6刺激B细胞分化以产生抗体，促进激活T细胞的扩增，扩大造血细胞产物生成，并诱导急性期蛋白的表达。

FUJIFILM Wako Shibayagi Corporation的LBIS Cytokine ELISA Kit系列推出了新产品Human IL-7。对于现有产品因缺乏灵敏度而无法检测的正常样品和低浓度样品，新产品可以高灵敏度且重复性良好地进行检测。

IL-7主要由骨髓、胸腺、淋巴器官或组织的基质细胞等非造血细胞产生，是诱导干细胞分化为免疫细胞的细胞因子。成熟体由152个氨基酸组成。CD4+、CD8+两种细胞的增殖和发生、促进抗病毒活性，还与类风湿关节炎、慢性结肠炎的产生以及与嗜酸性粒细胞活性引起的哮喘有关。另外，作为抗癌药和免疫检查点相关药物联合治疗中的助推因子，高灵敏度IL-7的检测，在NK细胞、B细胞和T细胞的增强作用物质的探索和研究中也备受期待。
