应用背景:

当用HIS3作为报告基因的酵母杂交筛选中,HIS3会有一定程度的泄露表达,导致背景过高的发生,此时需在筛选培养基内加入HIS3酶竞争性抑制剂——3-AT(MX7458)降低背景表达。需注意的是,加入过高浓度的3-AT,蛋白间存在较弱的相互作用会被抑制,因此,强烈建议针对自身系统确定最佳的工作浓度。

在正式进行酵母单杂或双杂筛选,就诱饵质粒(蛋白)进行自激活和HIS3报告基因基本表达水平的确定,这一实验用来确定诱饵是否存在激活转录和自激活是否能被3-AT所抑制。3-AT是HIS3酶的竞争性抑制剂,用来滴定组氨酸缺陷培养基上生长时HIS3基因的最低表达水平。
