酵母缺陷型培养基是由SD Base + DO Supplement,以添加琼脂和未添加琼脂两种方式混合而成的完全培养基,前者是固体培养基,后者是液体培养基(肉汤),用于选择培养含特异载体的酵母或进行杂交筛选。
Minimal SD Base,即Minimal Synthetical Defined Medium,是在无氨基酸酵母氮源基础(YNB)上添加单一碳源如葡萄糖、半乳糖、棉子糖和琥珀酸盐或多碳源搭配而合成的培养基基础。Dropout (DO) Supplement,即氨基酸缺陷混合物(或省却成分添加剂)是缺乏一种或多种特定氨基酸的混合物。
SD/-His/-Leu/-Trp/-Ura with Agar就是缺乏组氨酸(His),亮氨酸(Leu),色氨酸(Trp)和尿嘧啶(Ura)四种成分的酵母缺陷型固体培养基,常用于双质粒酵母转化筛选和双/三杂交系统报告基因检测。
保存与运输方法
保存:室温干燥保存,至少2年稳定。
运输:室温运输。
培养基配制
1) 取一包粉末,全部加入500ml去离子水,搅拌混匀使其溶解。琼脂不能溶解直至高压灭菌后。
2) 重要:测试培养基pH是否为5.8,若有必要用弱酸或弱碱调整pH。
3) 121℃高压灭菌15min。冷却至~50℃后再倒板。
4) 室温下使得平板培养基凝固,用封口膜封好后倒置保存在4℃。

Minimal SD Base,即Minimal Synthetical Defined Medium,是在无氨基酸酵母氮源基础(YNB)上添加单一碳源如葡萄糖、半乳糖、棉子糖和琥珀酸盐或多碳源搭配而合成的培养基基础。Dropout (DO) Supplement,即氨基酸缺陷混合物(或省却成分添加剂)是缺乏一种或多种特定氨基酸的混合物。
SD/-Ade/-His/-Leu/-Trp with Agar就是缺乏腺嘌呤(Ade),组氨酸(His),亮氨酸(Leu)和色氨酸(Trp)四种成分的酵母缺陷型固体培养基,常用于双质粒酵母转化筛选和双/三杂交系统报告基因检测。
保存与运输方法
保存:室温干燥保存,至少2年稳定。
运输:室温运输。
培养基配制
1) 取一包粉末,全部加入500ml去离子水,搅拌混匀使其溶解。琼脂不能溶解直至高压灭菌后。
2) 重要:测试培养基pH是否为5.8,若有必要用弱酸或弱碱调整pH。
3) 121℃高压灭菌15min。冷却至~50℃后再倒板。
4) 室温下使得平板培养基凝固,用封口膜封好后倒置保存在4℃。

高盐察氏培养基(Salt Czapek Dox Agar),又称为高盐察氏琼脂、高盐察氏琼脂培养基、高盐查氏培养基,用于霉菌和酵母菌的计数。以硝酸钠为唯一氮源,葡萄糖为碳源。磷酸氢二钾提供缓冲力,硫酸镁、氯化钾、硫酸亚铁提供必需的离子。高浓度的氯化钠减缓细菌生长速度。琼脂用作凝固剂。

酵母缺陷型培养基是由SD Base + DO Supplement,以添加琼脂和未添加琼脂两种方式混合而成的完全培养基,前者是固体培养基,后者是液体培养基(肉汤),用于选择培养含特异载体的酵母或进行杂交筛选。
Minimal SD Base,即Minimal Synthetical Defined Medium,是在无氨基酸酵母氮源基础(YNB)上添加单一碳源如葡萄糖、半乳糖、棉子糖和琥珀酸盐或多碳源搭配而合成的培养基基础。Dropout (DO) Supplement,即氨基酸缺陷混合物(或省却成分添加剂)是缺乏一种或多种特定氨基酸的混合物。
SD/-Trp/-Ura Broth就是缺乏尿嘧啶(Ura)和色氨酸(Trp)两种成分的酵母缺陷型液体培养基,主要用于双质粒酵母转化筛选和单/双杂交系统报告基因检测。
保存与运输方法
保存:室温干燥保存,至少2年稳定。
运输:室温运输。
培养基配制
1) 称取28g粉末加入1L去离子水,搅拌混匀使其溶解。
2) 重要:测试培养基pH是否为5.8,若有必要用弱酸或弱碱调整pH。
3) 121℃高压灭菌15min。4℃避光保存已灭菌的液体培养基。

4) 室温下使得平板培养基凝固,用封口膜封好后倒置保存在4℃。

产品描述
酵母缺陷型培养基是由SD Base + DO Supplement,以添加琼脂和未添加琼脂两种方式混合而成的完全培养基,前者是固体培养基,后者是液体培养基(肉汤),用于选择培养含特异载体的酵母或进行杂交筛选。
Minimal SD Base,即Minimal Synthetical Defined Medium,是在无氨基酸酵母氮源基础(YNB)上添加单一碳源如葡萄糖、半乳糖、棉子糖和琥珀酸盐或多碳源搭配而合成的培养基基础。Dropout (DO) Supplement,即氨基酸缺陷混合物(或省却成分添加剂)是缺乏一种或多种特定氨基酸的混合物。
SD/-Leu/-Trp Broth就是缺乏亮氨酸(Leu)和色氨酸(Trp)两种成分的酵母缺陷型液体培养基,主要用于双质粒酵母转化筛选和单/双杂交系统报告基因检测。
保存与运输方法
保存:室温干燥保存,至少2年稳定。
运输:室温运输。
培养基配制
1) 称取28g粉末加入1L去离子水,搅拌混匀使其溶解。
2) 重要:测试培养基pH是否为5.8,若有必要用弱酸或弱碱调整pH。
3) 121℃高压灭菌15min。4℃避光保存已灭菌的液体培养基。

运输:室温运输。
培养基配制
1) 取一包粉末,全部加入500ml去离子水,搅拌混匀使其溶解。琼脂不能溶解直至高压灭菌后。
2) 重要:测试培养基pH是否为5.8,若有必要用弱酸或弱碱调整pH。
3) 121℃高压灭菌15min。冷却至~50℃后再倒板。
4) 室温下使得平板培养基凝固,用封口膜封好后倒置保存在4℃。

SD/-His/-Leu with Agar就是缺乏组氨酸(His)和亮氨酸(Leu)两种成分的酵母缺陷型固体培养基,常用于双质粒酵母转化筛选和单/双杂交系统报告基因检测。
保存与运输方法
保存:室温干燥保存,至少2年稳定。
运输:室温运输。
培养基配制
1) 取一包粉末,全部加入500ml去离子水,搅拌混匀使其溶解。琼脂不能溶解直至高压灭菌后。
2) 重要:测试培养基pH是否为5.8,若有必要用弱酸或弱碱调整pH。
3) 121℃高压灭菌15min。冷却至~50℃后再倒板。
4) 室温下使得平板培养基凝固,用封口膜封好后倒置保存在4℃。

背景介绍
察氏琼脂(Czapek Dox Agar)是根据Thom&Church研发的配方制备的培养基,具限定性化学成分。含硝酸氮用作唯一氮源的一种半合成培养基。广泛用作真菌常规培养的培养基之一。只有非苛养的土壤细菌才能生长。察氏琼脂能用于白色念珠菌的厚垣孢子产生试验。因含不同盐成分具良好的缓冲能力。pH值略高于中性。察氏琼脂支持几乎所有腐生曲霉(具菌丝体和分生孢子形成特征)的大量生长。
根据Raper&Fenell (1965)的报道,添加1%玉米浆有助于绝大多数曲霉属的生长和孢子产生。按照5g/L加入酵母粉以及pH=4.0推荐用于分离土壤真菌。加入30mg/L链霉菌素和2mg/L金霉素能抑制可能存在的细菌菌丛生长。

产品用法
称取本品50.0g,加入1L蒸馏水(或去离子水)中,加热煮沸至完全溶解,分装,121℃高压灭菌20min。倒板前请摇匀。

保存:室温密封阴凉干燥处避光保存,3年有效。
运输:室温运输。
微生物灵敏度实验
根据产品用法配制的培养基,接种如下质控菌株,置于25℃培养箱、培养3~5天。

Agar, Gel Strength 900~1200 g/cm2, 琼脂粉 植物研究中常用琼脂粉为凝固基质
琼脂粉一般是从江蓠、紫菜、石花菜中制取的,可用作增稠剂、凝固剂、悬浮剂、乳化剂、稳定剂、保鲜剂、粘合剂和生物培养基。
微生物研究中常用琼脂粉为凝固基质,用来配制各种微生物固体和半固体培养基,比如LB固体培养基,YNB固体培养基,PDA培养基。相较于更传统的琼脂条作为凝固基质,溶解性强,培养基透明度大大提高,利于后续的微生物菌落研究。
植物研究中常用琼脂粉为凝固基质,用来培养各种植物研究材料如水稻,玉米,拟南芥,花卉等培养基的凝固剂,比如MS,B5,N6固体培养基等。
本品凝胶强度900~1200 g/cm2,用作微生物研究和植物组培原料,一般添加量1.5~2.0%。

注意事项
1) 琼脂粉默认使用pH值为中性,当pH<4无法凝结。
2) 琼脂粉使用过程遇到不凝固,需要考虑以下因素:①培养基pH值过低,偏酸导致水解和不凝;②所用琼脂量不够,增加用量;倒板前未充分混匀,凝固不均匀;③灭菌温度过高,加热时间过长,pH变化或破坏了琼脂的凝固度;④培养基加热溶解不充分,导致没有凝固。
