Description

The Vial Clamp™ – Straight is a chrome plated, hemostat style tool. It has a tip shaped to hold the storage vial of all the CrystalCap Systems™ straight when the clamp is closed. The clamp can be locked using the hemostat style closure. This clamp makes it easy to dip the storage vial into liquid nitrogen for crystal storage. The complete length of the clamp is straight, with an overall length of 195 mm. When the vial is placed in the clamp, the length of it is positioned perpendicular, 90° to the length of the clamp.

Description

The Vial Clamp – Curved is a chrome plated, hemostat style tool. It has a tip shaped to hold the storage vial of all the CrystalCap Systems at an angle when the clamp is closed. The clamp can be locked using the hemostat style closure. The end of the clamp, where the vial is held, is curved at either a 45°/135° or 110°/ 70° angle. When the vial is placed in the clamp, the length of it is positioned to the clamp at either of those same angles. The overall length of each clamp is the same, 195 mm.

Description

18 mm Mounted CryoLoops with 20 micron diameter nylon. Nylon loops are staked to hollow, stainless steel MicroTubes™ that are used to mount, freeze, and secure the crystal during cryocrystallographic procedures and X-ray data collection. The MicroTube on the 18 mm Mounted CryoLoops is 18 mm in length and ready to insert into CrystalCap Magnetic and CrystalCap HT. The 20 micron diameter nylon loops show minimal diffraction, are thin for fast cryo cooling of the crystal, strong, and aerodynamic.

For use with CrystalCap, CrystalCap ALS, CrystalCap ALS HT and CrystalCap SPINE HT systems. Not compatible with the CrystalCap Copper, CrystalCap Copper HT, or CrystalCap ALS HT.

To obtain the desired length of MicroTube, simply snap the MicroTube at the desired length and secure to the CrystalCap using epoxy or similar adhesive. These nylon loops show minimal diffraction, are thin for fast freezing, strong, and aerodynamic..

Most data collection geometries prefer or require a 22 mm length between the base of the cap and the beam. For a 22 mm overall length using the CrystalCap, CrystalCap ALS, CrystalCap ALS HT and CrystalCap SPINE HT, snap the MicroTube at the second notch from the bottom, creating an 18 mm MicroTube lenghth and secure to the CrystalCap using epoxy or similar adhesive. Description

Similar to the Mounted CryoLoop™ in design and function, the Adjustable Mounted CryoLoop allows one to manipulate and orient the CryoLoop to multiple, unique positions previously unattainable with typical cryo setups. The Adjustable Mounted CryoLoop is a 20 micron diameter synthetic loop mounted inside a malleable stainless steel sleeve, which in turn, is mounted inside the EasySnap MicroTube™.

Using forceps or a probe one can manipulate the angle and orientation of the CryoLoop by bending and adjusting the angle of the malleable stainless steel insert which holds the CryoLoop. The Adjustable Mounted CryoLoop may be positioned several times. Note: Repeated manipulation will fatigue the sleeve which can lead to failure of the sleeve. The Adjustable Mounted CryoLoop is designed to work with the CrystalCap System components.

The MicroTube is 24 mm in length and is specially engineered with EasySnap™ notches at the 10, 12, 14, 18, and 21 mm measures. Double/Bubble® Epoxy HR4-347 extra fast setting with a work time of 3 to 5 minutes. The preferred epoxy for cryocrystallography. Sold as a pack of 5. Hardman Double/Bubble Fast-Setting Epoxy is an extra fast setting, two part, room temperature curing epoxy adhesive. It has a convenient mix ratio, excellent cohesive strength and bonds to most substrates including metal, wood, concrete, ceramics and many plastics. The Double/Bubble package is a handy, dual-pouch, one-shot, job-size packaging concept. Fills gaps, will not shrink. Contains no solvents. Two part epoxy in a single, dual-pouch 3.5 gram packet.

To use:
1. Fold Double/Bubble packet
2. Cut or tear to open end
3. Squeeze out adhesive
4. Mix parts A and B together with supplied spatula

Description

The CrystalCap is a magnetic sample mount (also known as a cap, pin or goniometer base) designed for cryocrystallography systems that accept SSRL style sample mounts. The CrystalCap attaches to a magnetic CrystalCap Vial and Magnetic Goniometer Base. The tip of the CrystalCap accepts a Mounted CryoLoop™.

The CrystalCap Vial is a 1.8 ml cryo vial featuring two small vents. A ring magnet is molded into the open end of the vial so that when the cap is positioned in the vial, the ring magnet holds the cap on the vial during cryogenic storage.The HR4-904 CrystalCap Vial does not have a magnet on the bottom of the vial.

The conical shape with ledge is compatible with SSRL style grippers, auto mounters and sample handlers.

Description

The CrystalCap ALS HT is a magnetic sample mount (also known as a cap, pin or goniometer base) designed for cryocrystallography systems that accept ALS style sample mounts. The CrystalCap ALS HT attaches to a magnetic CrystalCap Vial and magnetic goniometer base. The tip of the CrystalCap accepts a Mounted CryoLoop™.

The CrystalCap Vial is a 1.8 ml cryo vial featuring two small vents. A ring magnet is molded into the open end of the vial so that when the cap is positioned in the vial, the ring magnet holds the cap on the vial during cryogenic storage. The HR4-904 CrystalCap Vial does not have a magnet on the bottom of the vial.

The ledge free conical shape of the CrystalCap ALS HT is compatible with ALS style grippers, auto mounters and sample handlers.

The CrystalCap ALS HT features a two dimensional (2D) alphanumeric 16 x 16 data matrix code on the underside of the cap. Each cap is also color coded for CryoLoop size.

Note: CrystalCap ALS HT with Mounted CryoLoops are sold without vials. Vials HR4-904 sold separately.

Description

The CrystalCap SPINE HT is a magnetic sample mount (also known as a cap, pin or goniometer base) designed for cryocrystallography systems that accept SPINE style caps. The CrystalCap SPINE HT attaches to a magnetic CrystalCap SPINE Vial and magnetic goniometer base. The tip of the CrystalCap SPINE HT accepts a Mounted CryoLoop™.

The CrystalCap SPINE Vial is a 1.8 ml cryo vial featuring two small vents and is compatible with the CrystalCap SPINE HT. A ring magnet is molded into the open end of the vial so that when the cap is positioned in the vial, the ring magnet holds the cap on the vial during cryogenic storage. The CrystalCap SPINE Vial features chamfered edges for enhanced cap positioning as well as a magnetic alloy bottom for stability. The HR8-114 CrystalCap SPINE Vial does have a magnet on the bottom of the vial.

The hollowed out design of the CrystalCap SPINE HT is compatible with SPINE style grippers, auto mounters and sample handlers.

The CrystalCap SPINE HT features a two dimensional (2D) alphanumeric 16 x 16 data matrix code on the underside of the cap. Each cap is also color coded for CryoLoop size.

CrystalCap SPINE without 2D code/printing (HR8-094) is the CrystalCap SPINE HT cap only, without 2D code, without color code, without white white background on bottom of cap and no alphanumeric side labeling; cap only, without Mounted CryoLoop.

Note: Caps with Mounted CryoLoops are sold without vials. Vials available separately.

The Magnetic Base (9.3 mm diameter support) is a machined brass piece fitted with a magnet. The base fits into the goniometer head, providing a secure base for the CrystalCap System™ sample holders. Description

The Crystal Crusher is designed to crush macro crystals into micro crystals for seeding and other applications. The hemispherical end of the tool fits round, concave bottomed sitting drop crystallization plates for efficient crystal crushing. The Crystal Crusher is molded from solid, borosilicate glass. One end of the tool features a larger diameter, cylindrical handle, while the opposite end features a smooth, hemispherical end for crushing crystals without damaging crystallization plates.

Description

The Seed Bead kits are used to create a seed stock for performing subsequent seeding experiments. The Seed Bead kits contains 24 Seed Bead Tubes, a special 1.5 ml microcentrifuge tube, shaped to maintain the seed in the sample during mixing. Each tube is configured with a PTFE, Stainless Steel, Zirconium Silicate Ceramic, or Glass Seed Beads.

Seeding allows one to grow crystals in the metastable zone. Crystallization in this zone provides control, reproducibility and an improved likelihood of a successful crystallization experiment. Also, crystals can grow from seeds but cannot spontaneously nucleate. By placing a seed or solution of seeds in a drop which is saturated to the metastable zone, one can use the seeds to grow larger single crystals. By controlling the number of seeds introduced into the drop, one can control the number of crystals grown. It is not practically possible to measure and know the number of seeds introduced to a drop, but by performing serial dilutions from a concentrated seed stock, one can control the number of crystals grown in the drop.

Using the Seed Bead kits, one can create crystal seed stock for subsequent seeding experiments. Crystals are placed in the microcentrifuge tube with the Seed Bead(s) and either vortexed or sonicated to generate a seed stock. Then by performing serial dilutions, one can control the number and size of crystals in the experiment.

The Seed Bead kit is useful for the preparation of seed stocks for automated and semi-automated microseeding (D’Arcy 2007, Harlos 2008).

Each Seed Bead Kit (HR2-320) contains 24 special microcentrifuge tubes, each tube containing a single 3mm PTFE Seed Bead.

Each Seed Bead Steel Kit (HR4-780) contains 24 special microcentrifuge tubes, each tube containing a six 1.6 mm Stainless Steel Seed Beads.

Each Seed Bead Ceramic Kit (HR4-781) contains 24 special microcentrifuge tubes, each tube containing a ten 1.0 mm Zirconium Silicate Ceramic Seed Beads.

Each Seed Bead Glass Kit (HR4-782) contains 24 special microcentrifuge tubes, each tube containing a ten 1.0 mm Glass Seed Beads.

Description

The LCP Sandwich Set consists of a base glass slide and an optimized cover slip. LCP Sandwich Set developed jointly with the renowned Scripps Research Institute in La Jolla, California, USA and is manufactured by Paul Marienfeld (0890003).

The LCP Glass Sandwich Set is a specially designed plate for either automated or manual setting of 96 Lipidic Cubic Phase matrix screening experiments. The plate can also be used for the bicelle method and batch method. The thin (< 2 mm high) plates have exquisite optical properties and are well suited for the detection of microcrystals and for birefringence-free imaging between crossed polarizers. The plates allow for in meso crystallization trials with 50 nanoliters protein/lipid mesophase and 1 microliter precipitant solution per trial.

The 96 well LCP Glass Sandwich Plate consists of a) 127.8 x 85.5 x 1 mm glass base plate with the footprint of an SBS-compliant plate, a 140 µm thick double sticky spacer with 96 punched out holes and b) a 0.2 mm thick glass coverslip. The double sticky spacer is already adhered to the base plate. A brown paper liner covers and protects the top of the double sticky spacer and base plate. The 0.2 mm thick glass coverslip fits over and seals the entire 96 well lower plate. Alternatively a series of twenty-four siliconized 18 x 18 mm No. 1 square cover slides (HR3-152) can be used to seal the entire 96 well lower plate. Each 18 x 18 mm No. 1 cover slide seals 4 wells. Space for a bar code is available at the left end of the plate. Each well can contain 50 nanoliters of cubic phase and 1 microliter of crystallization reagent.

18 x 18 mm No. 1 Siliconized Cover Slides are made from chemically resistant borosilicate glass D 263 M of the first hydrolytic class. The slides are colorless, clear, and suitable for fluorescence microscopy. The slides feature a super hydrophobic surface on both sides. The slides are No. thickness (0.13 ro 0.16 mm). The slides are supplied in a two compartment plastic box, 100 slides per box, 10 boxes per case, 1,000 slides total for HR3-152.

The Tungsten-carbide glass cutter can be used for cutting and removal of the upper coverglass that seals the LCP Sandwich Set. Sold separately.

Description

FRAP (Fluorescence recovery after photobleaching) measures membrane protein mobility in lipids in a high throughput fashion. The system utilizes an automated high throughput LCP-FRAP assay to prescreen membrane protein crystallization conditions.

The FRAP assay can be used to pre-screen multiple protein constructs, lipids and additives to eliminate those conditions that are non-conducive to protein diffusion, and thus crystallization, from the subsequent crystallization trials. High protein mobility and fast diffusion rates correlate well with crystallization conditions.

The FRAP sandwich set consists of a base glass slide with sealing film and an optimized cover slip. This product has been developed jointly with the renowned Scripps Research Institute in La Jolla, California, USA.

The FRAP Sandwich Set is optimized for the pre-crystallization study of membrane proteins.

Developed at the NIH Roadmap Center for membrane protein research and routinely used in GPCR structural biology.

Bottom Slide Features
• Dimensions: approx 127.8 x 85.5 mm, thickness approx. 1 mm
• Superhydrophobic glass surface
• Covered with a 0.06 mm (60 micron) high spacer. This spacer has 96 recesses of 5 mm diameter. They are consistent with the SBS format for robotic handling. The spacer has an adhesive surface to which the hydrophobic coverslip attaches. The crystallization drops are, hence, sealed in these reaction chambers.
