使用了多孔性填料的Q型,S型和M型相比于非多孔性填料可以承载更多蛋白质,即使一次大量进样峰形依然尖锐。所以此类型色谱柱适用于蛋白质的净化提纯。

应用数据

通常使用1-2 mol/l的(NH4)2SO4高浓度盐缓冲液作为初期流动相将样品吸附至弱疏水性固定相。之后采用逐步降低盐浓度的方法进行洗脱。下图的应用数据中显示了在高浓度盐溶液下,肌红蛋白(myoglobin)先于BSA被洗脱,证明了肌红蛋白(myoglobin)的疏水性低于BSA。

开放式柱色谱法的传统C18填料在30-50%的水浓度溶剂下性能就会受到很大限制。COSMOSIL (COSMOSIL) C18-OPN填料在硅胶表面键合了亲水基团从而增强了亲水性,可在100%水环境下使用。

Cosmosil色谱柱 (COSMOSIL) C18-OPN

C18-OPN填料在硅胶表面键合了亲水基,在硅胶孔内键合了十八烷基。此特性使得反相开放式柱色谱法可在100%水环境下使用。

可可碱 (Theobromine) 和茶碱 (Theophylline) 的分离

从结果可以看出样品在70%水环境下被很好的分离。

由于反相色谱法一般使用甲醇或水等高粘度溶剂,所以其流速低于使用低粘度溶剂的正相色谱法。另外,反相色谱法根据溶剂不同,流速也会有很大变化。右图显示了流速与溶剂组成的关系。可以看出高流速型140 um的流速是高分离型75 um的约2.5倍。

与正相色谱的比较

p-Cresol 和 p-Ethyphenol在结构上只有一个亚甲基的差别,它们在正相下很难被分离。然而使用C18-OPN可以很容易的将它们完全分离。

填料的再现性

可使用四氢呋喃,氯仿等溶剂冲洗填料。C18-OPN填料具有很好的再现性,可以重复使用。

“注意”

不要使用pH大于7的碱性溶剂或pH小于2的酸性溶剂冲洗填料。碱性过强的溶液会溶解硅胶,酸性过强的溶液会使固定相脱落。请在50C以下干燥填料。

硅胶在物理性质上是溶于碱性溶液的。虽然COSMOSIL C18-MS-II柱是以硅胶为基础填料的,但是因为采用了*的封尾技术因此,即使在碱性条件下也可以在某种程度上使用。但是请注意根据甲醇等有机溶剂以及缓冲液的种类及浓度的不同,其耐久性会发生变化。因此为了使柱子的寿命更长请在pH7.5以下使用。

COSMOSIL Sugar-D填料为5μm的高纯多孔球形硅胶,对单糖和寡糖的分离效果很好,峰形尖锐,没有拖尾,柱寿命长。以100%水做流动相,COSMOSIL Sugra-D的保留时间小。COSMOSIL Sugar-D更有利于分离在传统的ODS固定相上没有保留的高亲水化合物。在分析单糖和低聚糖时,常用的丙氨基固定相的氨基液相色谱柱对糖有一定的吸附且柱寿命较短。这些问题在Sugar-D上都得到了解决。新的COSMOSIL Sugar-D柱耐久性能得到改善,而且还能得到比较好的(尖锐的)色谱峰。

由于糖类大都是低疏水性化合物,因此在普通的C18色谱柱均不能保留。 COSMOSIL Sugar-D和NH2-MS色谱柱是专为分离糖而设计的。对于疏水性的糖苷或糖类衍生物推荐使用COSMOSIL C18-PAQ色谱柱。

传统的氨丙基柱相比于Sugar-D具有较长的保留时间,Sugar-D的保留时间可通过在增加流动相中乙腈的浓度(5%~10%)作调整。
         常规氨丙基键合固定相用于高效液相色谱分析单糖和低聚糖。常规氨丙基键合固定相的一般缺点是存在拖尾现象,且容易吸附某些糖和一般耐久性较低(使用寿命短)。新型COSMOSIL Sugar-D柱很好地解决了这些问题,能够更好地分离糖类,且耐用性大大提高,且拥有优越的分离性能和高回收率。

COSMOSIL 5C18-MS-II 

COSMOSIL 5C18-MS-II 液相色谱柱采用新的封端技术,可代替 COSMOSIL 5C18-MS 色谱柱。由于流动相的强极性组分和硅胶填料的化学性质,极性基体封端硅胶可产生屏蔽效应。因此采用新的封端技术分离碱性化合物时峰形尖锐,硅胶的吸附量小,适合分离螯合化合物。 5C18-MS-II 另一个优点是与 5C18-MS 相比保留时间小, 5C18-MS-II 更合适于 HPLC 条件。

OSMOSIL HILIC色谱柱

COSMOSIL HILIC是新型的亲水相互作用色谱柱,三唑键合硅胶。亲水相互作用色谱法是一种变异的正相色谱,极性固定相,流动相含有高浓度的有机溶剂和低浓度的水溶液淋洗液。主要分离机制是在极性固定相和非极性流动相之间对极性组分的分离。也叫做水相正相体系。淋洗顺序类似于正相体系,样品淋洗是为了增加亲水性。 COSMOSIL HILIC不使用离子对试剂,对在反相色谱中没有保留的极性组分保留强。同时带正电荷的固定相也体现阴离子离子交换色谱机制,对酸性化合物保留强。

Sugar-D

以往单糖和低聚糖都是用氨丙基键合固定相的液相色谱分析,这种柱子的缺点是峰形拖尾、柱寿命短、对某些特定的糖吸附小。但是 COSMOSIL® Sugar-D 解决了这些问题,分离效果更好,峰形尖锐,增加了柱寿命。以 100 %水做流动相, COSMOSIL ® Sugar-D 的保留时间小。对比传统的 C18 柱, COSMOSIL® Sugar-D 更有利于分离高亲水化合物。

     氨丙基柱对某些特定的糖如树胶醛糖或半乳糖等只有部分吸附,样品消失或峰形拖尾,COSMOSIL® Sugar-D 分析这些糖 能得到尖锐峰形。
