【使用目的】
耐熱性好酸性菌Alicyclobacillus acidoterrestrisの鑑別
【操作方法】
1.測定操作法
(1) Va‐YSG培地にYSG平板培地で分離した芽胞を形成
していない新鮮コロニーを10μL白金耳にて1白金耳
懸濁させて下さい。（1×108cfu/mL 以上）
(2) 45±1℃の孵卵器にて 3 時間インキュベートして下さ
い。
(3) 試液１を 1.0mL 加えて下さい。
(4) 試液２を 20μL 加えて下さい。
(5) 試液３を２0μL 加えて下さい。
(6) 十分混合して下さい｡
(7) 5～10 分、室温に静置して下さい。
(8) 目視にて反応液の色の変化を確認して下さい。
※目視判定は 10 分以内に行なって下さい。
2.ブランクの調製
菌を接種しない Va‐YSG 培地をブランクとして検体と
同様に(2)～(8)の手順で試験して下さい。
3.陽性コントロールの調製
精製水 2mL に陽性コントロール 100μL を加えて撹拌
した後、検体と同様に(３)～(8)の手順で試験して下さい。

【使用目的】
耐热嗜酸性菌Alicyclobacillus acidoterrestris的鉴别
【操作方法】
1.测量操作法
(1)在Va – YSG培养基上用YSG平板培养基分离形成芽胞
将新鲜菌落10μL白金耳加1白金耳
请让我悬而未决。(1×108cfu/mL以上)
(2)在45±1℃的孵化器中孵化3小时。

(3)请加入1.0mL试液1。
请加入20μL试液2。
请加入20 μL试液3。
(6)充分混合。
(7)在室温下静置5 ~ 10分钟。
(8)请用目视确认反应液的颜色变化。
※目视判定请在10分钟以内进行。
2.毛坯的制备
没有接种细菌的Va – YSG培养基，
同样请按照(2)~(8)的顺序进行测试。
3.阳性控件的制备
在2ml纯净水中加入100μL阳性控浆搅拌
之后，请和检体一样按照(3)~(8)的顺序进行试验。
